      将40只健康的SD大鼠,随机分为2型糖尿病(type 2 diabetes mellitus,T2DM)实验组和对照组,T2DM实验组利用 STZ链脲佐菌素 (STZ)小剂量分次给药进行T2DM造模,然后喂食高脂、高糖和高蛋白饲料8周。采用Morris水迷宫法评估T2DM组大鼠和对照组大鼠的认知功能,研究其与双侧海马1H-MRS(NAA、 Cho、 Cr、mI代谢物曲线下面积及其代谢物的比值)之间的相关性,并采用HE染色检测海马组织中是否有淀粉样蛋白沉积。结果发现T2DM实验组大鼠的学习潜伏期较对照组延长,与左侧海马NAA值减低呈负相关,记忆潜伏期较对照组缩短,与左侧海马NAA值减低呈正相关, (1H-MRS)中的双侧海马NAA值较对照组均减低,以左侧为著,差异均有统计学意义(P<0.05)。HE染色可见T2DM实验组大鼠脑组织有明显的淀粉样物质沉着,对照组大鼠海马组织中的淀粉样物质沉积不明显。研究表明T2DM引起大鼠海马组织学变化及代谢异常并损害认知功能,1H-MRS可以为诊断T2DM脑损伤和评估认知功能提供简便有效的影像学方法与策略。
英文摘要:
      Forty Sprague Dawley (SD) healthy rats were randomly divided into type 2 diabetes mellitus (T2DM) group and control group. Rat models of T2DM were established by intraperitoneal injection of streptozocin (STZ), and then the rats were fed on high-fat, high-sugar and high-protein for 8 weeks. The cognitive function was evaluated by Morris water maze,to determine the effect of T2DM on spatial learning and memory in rats, and investigate the correlation between cognitive function and 1H-MRS(NAA, Cr, Cho and mI,and their ratios were assessed by statistical t test) on bilateral hippocampus. HE staining was used to observe the deposition of amyloid in hippocampus. The T2DM experimental group had a longer earning latency and was negatively correlated with the decrease of NAA in the left hippocampus (p<0.05), it is a shorter memory latency than the control group and positively correlated with the decrease of NAA in the left hippocampus (p<0.05). The differences of NAA between the T2DM experimental group and the control group were statistically significant (p<0.05) with more obvious impairment in left hippocampus (p<0.05), and a large number of amyloid structures were observed under HE staining. T2DM caused histological changes and metabolic abnormalities in hippocampus of rats and impaired cognitive function, (1H-MRS)may provide effective imaging strategies and methods to early diagnosis brain damage and quantitative assessment cognitive function in T2DM.
